New study finds intermittent fasting may ease depression by rewiring the brain’s dopamine system.

Intermittent fasting has already earned a reputation for helping with weight and metabolism, but new research suggests it might also lift your mood by directly targeting the brain’s dopamine pathways. A study from the Second Hospital of Jilin University, published in Neurobiology of Disease, explored how fasting could fight depression—and the results were pretty compelling, at least in mice.

Researchers used a classic mouse model of depression where animals are exposed to random stressors—like foot shocks or water deprivation—over several weeks. Some of these stressed mice then started an intermittent fasting routine, switching between 24 hours of food and 24 hours without. Others were given either a standard antidepressant (fluoxetine) or a shorter fasting window for comparison.

The mice on the 24-hour fasting plan showed a clear turnaround. They were more eager to drink sweetened water, a sign they could still enjoy simple pleasures, and they spent less time sitting still in tests of despair. When scientists dug into their brains, they found heightened activity in the medial prefrontal cortex (mPFC)—a region tied to emotion and decision-making. More importantly, fasting boosted signaling in a key dopamine D1 receptor pathway, which is known to regulate mood.

To prove this was the critical piece, the team blocked these dopamine receptors with a drug, wiping out fasting’s benefits. They also used optogenetics to directly stimulate the same brain cells, recreating the antidepressant effects even without fasting. While it’s still early days—this was all in mice—these findings hint that intermittent fasting might one day be a natural option to help manage depression by literally tuning up the brain’s mood circuits.
